In supplement form it is typically derived from bovine or porcine cartilage. It is used in joint support supplements primarily in combination with glucosamine. ## Mechanism of Action Chondroitin's primary mechanism is inhibition of cartilage-degrading enzymes including matrix metalloproteinases and aggrecanases, which break down the proteoglycan matrix of cartilage in osteoarthritic joints. It also inhibits the synthesis of inflammatory mediators in chondrocytes and may stimulate the synthesis of hyaluronic acid, a component of synovial fluid. The combination of glucosamine and chondroitin is considered to have complementary mechanisms: glucosamine provides the building blocks for cartilage synthesis while chondroitin inhibits the enzymes that degrade it. ## Evidence Summary Chondroitin shares the mixed evidence profile of glucosamine. Some trials show benefit, others show limited effect over placebo. The GAIT trial in humans (2006, New England Journal of Medicine) found that the glucosamine-chondroitin combination showed significant benefit in a subgroup of patients with moderate to severe pain, though not in the overall study population. Veterinary-specific evidence is limited. A 2003 study by Moreau et al in dogs found improvement in force plate measurements with glucosamine-chondroitin supplementation.
